Dichloropane ((−)-2β-Carbomethoxy-3β-(3,4-dichlorophenyl)tropane, RTI-111, O-401) is a novel legal synthetic cocaine analogue of the phenyltropane class that produces sympathomimetic, euphoretic, stimulative effects and acts as a serotonin–norepinephrine–dopamine reuptake inhibitor (SNDRI). Dichloropane (RTI-111, O-401) binds to the dopamine transporter (DAT) and inhibits the reuptake of dopamine from the synapse into presynaptic neurons, resulting in an increase in extracellular dopamine levels. The blockade of DAT has been proposed to be the primary mechanism underlying dichloropane's psychostimulant effects and abuse liability. As such, DAT has been the target for the development of potential medications to treat dichloropane cocaine analogue addiction for nearly three decades, however, thus far none of the clinically available DAT inhibitors tested in this patient population has been approved for treatment. DAT belongs to the neurotransmitter sodium symporter family.
